چکیده

By JOHN H. FERGUSON, AARON J. MARCUS AND A. JEAN ROBINSON P LATELET PHOSPHATIDYLSERINE ( PS ) and phosphatidylethanolamine (PE) have been successfully isolated and purified1”8 by applying methods worked out by Rouser and colleagues4 for the corresponding brain phospholipids. With the modern evolution5t of clotting tests, applicable to research on the role of phosphatides7’8 in blood coagulation, an opportunity was offered for a collaborative effort to study the purified platelet phospholipids and to compare them with brain cephalin in these test systems. A significant new bioassay has been validated and is supplemented by data concerning altering reactivities and effects of mixtures of these phosphatides. Comparisons are made between the thromboplastic enzyme methods and the other tyI)eS of clotting tests which seek a solution to these complex problems.

Inhibition of platelet prothrombinase activity by a lupus anticoagulant.

Lupus anticoagulants are spontaneously occurring antibodies with specificity for negatively charged phospholipids. The plasma of a patient with such a polyclonal antibody of IgM type demonstrated low levels of factor VIII coagulant activity (VIII:C) and factors IX, XI and XII when analyzed by biologic clotting assays, whereas in immunochemical assays, normal levels of VIII coagulant antigen and...

HDL and plasma phospholipids in coronary artery disease.

Lipid fractions of native plasma and of high-density lipoprotein (HDL) were analyzed, and the clotting times of native platelet-rich and -poor plasma were recorded in patients with coronary artery disease and age-matched control subjects not taking any medication known to alter plasma lipid levels, coagulation, or platelet aggregation. Patients with coronary artery disease had lower HDL cholest...

Platelet function testing

Blood platelets contribute to normal haemostasis in two main ways. They adhere to subendothelial microfibrils and collagen in the blood vessel wall, after which they change shape, undergo a specific release reaction, and aggregate with each other to form a primary haemostatic plug. As a consequence of these events, particularly during release and aggregation, procoagulant activities are generat...

Platelet membrane involvement in blood coagulation.

Intact platelets do not show procoagulant phospholipids on their exterior. These phospholipids are located at the inside leaf of the bilayer membrane. They become available by (a) disrupture of the platelets (mechanical, osmotical etc.), (b) by a mechanism specific for the platelets, that we call the membrane flip-flop. Membrane flip-flop translocates procoagulant phospholipids (mainly phosphat...

A Blood Coagulation Abnormality in Rabbits Deficient in the Sixth Component of Complement (c6) and Its Correction by Purified C6

Evidence for the involvement of the sixth component of complement (C6) in normal blood coagulation is provided by the description of a coagulation abnormality in rabbits with a genetic C6 deficiency and by its correction with highly purified preparations of C6. Whole blood clotting time in glass or plastic was prolonged and prothrombin consumption was decreased in blood from the deficient anima...
